What is Pranidipine ?
Calcium antagonists are widely used to treat angina, systemic hypertension, and other cardiovascular diseases. 1,4 dihydropyridine (DHP) calcium antagonists bind to specific receptors in cardiac muscle and smooth muscle to inhibit the influx of extracellular calcium when these muscles contract in response to stimulation. The original drug nifedipine of DHP only had short cardiovascular effect, and later some second and third generation DHPS with long effect of antihypertensive effect appeared.

pranidipine is an extremely long-acting DHP. The difference between it and nifedipine lies in its slow
acting but long acting time, and it is speculated that its binding characteristics with cardiovascular
system receptors are also significantly different from nifedipine. Pratipine (Pranidipine, 1), chemically
named 2, 6-dimethyl-1, 4-dihydropyridine-4 -(3-nitrophenyl)-3, 5-pyridine-methyl dicarboxylate cinnamyl
ester, is clinically used in the treatment of hypertension. Pratipine has calcium channel antagonism, which
can relieve and inhibit calcium channel and block calcium ion flow. It can inhibit the release of intracellular
calcium ions. It is a new and long-acting calcium antagonist of dihydropyridine with unique pharmacological
and pharmacodynamic advantages.
Since the 1990s, there have been many researches on its synthesis
methods at home and abroad, and many new synthesis routes and improved methods have been reported.
Current animal experiments have shown that compared with nifedipine, nicardipine and nirendipine equal
calcium antagonists, pratipine has stronger antihypertensive activity and more permanent and gentle
antihypertensive effect. It only needs to be taken once a day and is well tolerated by patients. Therefore,
Pratipine is a potential antihypertensive drug.
